Highlight #1: Folic Acid Supplementation Lowers Blood Arsenic by Increasing as Methylation (manuscript in Preparation)

چکیده

Chronic arsenic (As) exposure currently affects more than 100 million people worldwide. Methylation of ingested inorganic arsenic (InAs) to methylarsonic(MMA) and dimethylarsinic acids (DMA) relies on folate-dependent one carbon metabolism. In a recent, randomized, controlled trial, we analyzed total As and As metabolites in urine and showed that folic acid supplementation resulted in an increase in the proportion of total urinary As excreted as DMA (%uDMA) and a reduction in %uMMA and %uInAs. Since DMA has a shorter circulating half-life than other arsenic metabolites, we hypothesized that facilitation of As methylation via folic acid supplementation might lower total blood As (tbAs) concentrations. Methodological advances, using inductively coupled mass spectrometry with a dynamic reaction cell (ICP-MS-DRC) have permitted us to test this hypothesis by measuring total As and As metabolites in blood in 130 participants with low plasma folate (< 9 nmol/L) before and after 12 weeks of supplementation with folic acid (400 μg/d) or placebo. These methodological advances have also led to the discovery that blood As is a biomarker of As exposure that is directly associated with the risk for arsenic-induced skin lesions. Thus, lowering blood As with folic acid could have therapeutic potential to reduce the risk for arsenic-induced illnesses.

Low-Dose Creatine Supplementation Lowers Plasma Guanidinoacetate, but Not Plasma Homocysteine, in a Double-Blind, Randomized, Placebo-Controlled Trial.

BACKGROUND Creatine synthesis from guanidinoacetate consumes ~50% of s-adenosylmethionine (SAM)-derived methyl groups, accounting for an equivalent proportion of s-adenosylhomocysteine (SAH) and total homocysteine (tHcys) synthesis. Dietary creatine inhibits the synthesis of guanidinoacetate, thereby lowering plasma tHcys in rats. OBJECTIVE We tested the hypotheses that creatine supplementati...

Thalassemia Intermedia; Folic Acid and Vitamin B12 Supplementation. What We Know and What is Needed?

This study presented a mini review on folic acid deficiency and recommendations for its supplementation in thalassemia intermedia (TI). TI is a clinical condition which lies between thalassemia major and thalassemia minor. Although TI patients may not need regular blood transfusion, precise diagnosis and management are critical for the prevention of clinical complications and quality of life im...

Effects of Altered Maternal Folic Acid, Vitamin B12 and Docosahexaenoic Acid on Placental Global DNA Methylation Patterns in Wistar Rats

Potential adverse effects of excess maternal folic acid supplementation on a vegetarian population deficient in vitamin B(12) are poorly understood. We have previously shown in a rat model that maternal folic acid supplementation at marginal protein levels reduces brain omega-3 fatty acid levels in the adult offspring. We have also reported that reduced docosahexaenoic acid (DHA) levels may res...

Effects of Folic Acid Supplementation on Proteinuria in Type 2 Diabetic Patients with Overt Proteinuria

Objective: An association between high homocysteine levels and proteinuria in diabetic nephropathy has been shown. On the other hand, supplementation with folic acid lowers homocysteine concentration. We have studied the effects of folic acid supplementation on proteinuria in type 2 diabetic patients with overt proteinuria. Materials and Methods: Forty five (29 men and 16 women) type 2 diabe...

A Preventive Approach to Arsenic Toxicity: Testing Folic Acid in Bangladesh

12 weeks was associated with an average 14% reduction in blood arsenic levels. In this issue of EHP, members of that team report further evidence that FA supplementation may be an effective intervention for arsenic-exposed populations. The study was conducted as part of the Health Effects of Arsenic Longitudinal Study, a prospective cohort study in Araihazar, Bangladesh. In a 24-week randomized...
